Potential anti-diabetic and anti-oxidant activity of lactic acid bacteria isolated from Indonesian traditional fermented Buffalo milk (Dadih): In silico approach
Dadih is a fermented Indonesian cuisine that originated in West Sumatera. Secondary metabolites generated by lactic acid bacteria isolated from Dadih were shown to have anti-diabetic and antioxidant properties in prior research. The goal of this work was to discover the anti-oxidant and anti-diabetic agent generated by lactic acid bacteria’s ligand-receptor interaction in order to examine a strong anti-oxidant and anti-diabetic drug from Dadih utilizing an in-silico technique. Secondary metabolites were identified and purified using a two-step process that included ammonium sulfate precipitation and gel filtration chromatography. LC-MS analysis revealed 12 compounds that probably correlate to anti-oxidant and anti-diabetic activity. AutoDock Vina was used to assess each compound’s potential anti-oxidant and anti-diabetic activity against alpha glucosidase as an anti-diabetic agent and aldehyde oxygenase as an anti-oxidant agent. The DL-I-Phenylalanine compound exhibited the lowest binding affinity (-6,7 kcal/mol) towards aldehyde oxygenase and - 6,5 kcal/mol against alpha glucosidase, according to molecular docking simulations. In a computer simulation, we discovered that DL-I-Phenylalanine molecules had a stable interaction with both the blood glucose and free radical receptors.
