The invention provides an isolation system and an isolation method. The isolation system comprises an isolation room, wherein the isolation room is isolated from an external environment by a movable door; the movable door comprises a connecting part and a movable door plate; a protective main body is disposed inside the isolation room, and a main body made of a flexible material is hollow inside and suitable for entering of a human trunk; the main body comprises an opening which extends along a vertical direction; sealing parts are disposed at two sides of the opening, and seal connection is formed between the two sealing parts; the sealing parts are suitable for connecting connection parts of the movable door; a first extension part is suitable for entering of a human head, and the main body is connected to the first extension part; a transparent window is disposed on the first extension part and suitable for observation of the external environment; a second extension part which can be bent is suitable for entering of a human arm and a human hand, and the main body is connected to the second extension part; a third extension part is suitable for entering of a human leg and a humanfoot, and the main body is connected to the third extension part; and one end of a connecting pipe communicates with space, and the other end is applicable to communicating with a gas source. The system and method have the advantages of being safe and efficient in isolation, etc.
