The invention discloses an open caisson and a negative pressure sinking construction process thereof. The open caisson comprises a well body and a well lid, an inner cavity of the well body is divided into several compartments through vertical partition plates, air bags matched with a well mouth in a seal mode are fixed to the periphery of the well lid, exhaust holes and drainage holes are formed in the positions, corresponding to the compartments, of the well lid, valves are arranged at the exhaust holes, and the drainage holes are connected with a sewage pump through a drainage pipeline. The open caisson structure is prefabricated in a prefabrication factory, and the construction procedures of foundation bed riprapping, foundation bed tamping and foundation bed leveling are not carried out before the open caisson is arranged, so that the overwater construction procedures are reduced, potential safety hazards are reduced, and offshore cover plate cast-in-place concrete after the open caisson is arranged can be reduced; through the connection between the air bags and an upper cover plate, the overall closed space in the sinking process of the open caisson is achieved, and negative-pressure sinking of the open caisson structure is achieved through an air floating platform; and through the air flotation operation platform, negative pressure sinking of the open caisson structure is achieved, and the sinking speed of the open caisson and the height difference after the open caisson enters soil are well controlled.
