The invention relates to a stand column construction method in one-column one-pile construction. A tool column is coaxially assembled at the upper end of a stand column, a clinometer is arranged in the stand column, and at least part of the tool column is exposed out of the ground when the stand column is inserted into a pile hole to the elevation; the stand column is hoisted, first-time verticality adjusting is conducted, the reading of the clinometer when the stand column is perpendicular is recorded as an initial value, and the stand column is inserted into the pile hole to the elevation; and the tool column is adjusted to conduct second-time verticality adjusting on the stand column till the reading of the clinometer is consistent with the initial value, the tool column is horizontally moved to enable the center of the tool column to be consistent with the design center, and center positioning of the stand column is completed. According to the stand column construction method in one-column one-pile construction, in the construction process, the assembled tool column is used for being matched with a stand column holding and inserting device to conduct holding and inserting construction on the stand column, the hoisting difficulty of the stand column is greatly reduced, the verticality adjusting and center positioning processes of the stand column are simplified, the perpendicular precision and the center positioning precision of the inserted stand column are both remarkably improved, and the construction quality is greatly improved.
