The invention discloses a germplasm resource safe preservation device and preservation method which are used for realizing safe preservation of the germplasm resources, saving energy, reducing emission and facilitating sampling of the germplasm resources. The storage device comprises a storage chamber, a transport well, a lifting unit, an air inlet well, an exhaust well and a monitoring unit; the storage chamber is of a reinforced concrete structure and is built under the ground surface, and an operation room, a treatment room, a buffering room and a storage room which are sequentially arranged and are independent of one another are arranged in the storage chamber; the transport well is arranged on the right side of the storage chamber; the lifting unit is arranged in the transport well and can move up and down, and a storage bottle containing the germplasm resources is placed on the lifting unit on the ground surface; the air inlet well and the exhaust well are arranged on the left side of the storage chamber, the air inlet well is communicated with the buffer room, and the exhaust well is communicated with the storage room; and the monitoring unit comprises a temperature sensor and a humidity sensor which are arranged in the storage room. According to the device and method, the germplasm resources can be safely preserved.
