The invention belongs to the field of application of concrete admixtures, and discloses a preparation method of an anti-flocculation amphoteric polycarboxylate superplasticizer, which comprises the following steps: step 1, preparing a mixture A by using unsaturated polyether as a macromonomer; 2, dissolving a chain transfer agent in a reducing agent to obtain a mixture B; step 3, removing the solvent under reduced pressure through rotary evaporation to obtain a faint yellow transparent liquid; step 4, preparing a mixture C; step 5, dropwise adding the mixture B and the mixture C into the mixture A at the same time, and mechanically stirring for 2-6 hours to obtain a reactant of the anti-flocculation amphoteric water reducing agent; and 6, pouring the reactant into a beaker, cooling to room temperature, and adjusting the pH value to 6-8 by using a NaOH aqueous solution to obtain the anti-flocculation amphoteric water reducer. The anti-flocculation amphoteric water reducing agent is used in concrete, and the problems that the fluidity of the concrete is reduced and the slump loss is too large after machine-made sand containing a flocculating agent is added into the concrete can be solved.
