The invention relates to a pavement thickness detection system and method for highway engineering. The pavement thickness detection system comprises a frame body structure; the sampling mechanism is arranged on the frame body structure and comprises a lifting assembly and a rotary drilling assembly which are arranged in a linkage mode, the rotary drilling assembly is slidably sleeved with a sampler, the rotary drilling assembly can drive the sampler to rotate, and the lifting assembly can drive the sampler to move in the vertical direction of the space in the rotating process of the sampler; the guide assembly is connected with the lifting assembly and the sampling device and comprises a telescopic plate and a driving plate, an embedding groove is formed in the driving plate, a first pulley rotationally installed on the telescopic plate slides in the embedding groove, and after the lifting assembly drives the sampling device to ascend to be flush with the frame body structure, the first pulley is driven by the driving plate to rotate. The telescopic plate pulls the sampler to be staggered with a through hole formed in the frame body structure through rolling fit of a first pulley and an embedding groove, so that rapid hole taking is realized, and the difficulty of taking out a sample is reduced.
