The invention discloses a prefabricated box girder hoisting device which comprises a traveling crane, a rack is fixedly connected to the bottom of the traveling crane, a fixing plate is arranged at the bottom of the rack, a steel wire rope is fixedly connected to the bottom of the fixing plate, and a positioning clamping mechanism is arranged on the rack. The positioning clamping mechanism comprises two sets of moving platforms, transmission components in transmission connection with the two sets of moving platforms correspondingly, a first motor connected with the transmission components, a second motor connected to the moving platforms and a positioning clamping plate connected with the second motor, the positioning clamping plate is vertically arranged outside the rack, and meanwhile the invention further provides a corresponding construction method. The movable platform is arranged to drive the positioning clamping plate to quickly clamp or loosen the prefabricated box girder, so that the engineering efficiency is improved, and meanwhile, the second motor is arranged to drive the positioning clamping plate to move in the vertical direction, so that the posture of the prefabricated box girder can be properly and finely adjusted to guarantee balance; and meanwhile, when not working, the travelling crane can move upwards to be folded, collision interference in the travelling crane moving process is avoided, and safety is improved.
