The invention discloses a one-time sinking construction method for a large open caisson. The one-time sinking construction method comprises the following steps: constructing a cushion layer and building a brick blank film; manufacturing the open caisson section by section; breaking and removing a cushion layer and a brick blank film; excavating and sinking by a small excavator; the crawler-type grab bucket excavates soil and sinks; the telescopic boom excavator excavates soil and sinks; sucking sludge by air and sinking; and the open caisson is subjected to underwater bottom sealing. The problems that in an existing open caisson construction mode, sinking is not in place easily, and the deviation exceeds the allowable deviation after the open caisson is subjected to final sinking are solved.
