The invention relates to a connection sealing mechanism of a water inlet valve and a water drain valve. The connection sealing mechanism comprises an installation rubber cushion assembly which is stuffed into an installation hole in the bottom of a water tank, and a base substrate which is arranged at the bottom of the water inlet valve or the water drain valve, and is inserted into the installation rubber cushion assembly, wherein the installation rubber cushion assembly is in clearance fit or interference fit with the installation hole in the initial state, and after the base substrate is inserted into the installation rubber cushion assembly, the installation rubber cushion assembly is in interference fit with the installation hole so as to realize sealing. A sealed connection method realizing easy assembly and disassembly of the water inlet valve and the water drain valve comprises the following steps: inserting the installation rubber cushion assembly into the installation hole in the bottom of the water tank, so that the installation rubber cushion assembly is in clearance fit or interference fit with the installation hole in the initial state; and inserting the base substrate into the installation rubber cushion assembly so that the installation rubber cushion assembly expands towards the periphery, and is in interference fit with the installation hole, and sealing is realized. The water inlet valve or the water drain valve can be conveniently assembled and disassembled inside the water tank if the water inlet valve or the water drain valve needs to be maintained or replaced, and the water tank of a closestool does not need to be detached, so that replacement and maintenance are rapid.
