The invention discloses a connection structure of a stand column and a transverse beam. The connection structure comprises the transverse beam and the stand column provided with mounting profiled grooves, wherein the transverse beam and the stand column are closed sectional material components; rotating locking holes are formed in the two sides of the stand column; rotating locking connecting pieces are arranged inside the rotating locking holes, extend toward the two sides of the stand columns and are provided with extension bodies; and supporting parts for supporting the extension bodies are fixedly connected to the interiors of the mounting profiled grooves of the stand columns and are in pluggable connection with the transverse beam at the two sides of the stand column. The invention also discloses a connection method utilizing the connection structure of the stand column and the transverse beam. The connection structure of the stand column and the transverse beam and the connection method utilizing the connection structure of the stand column and the transverse beam have the beneficial effects that installation procedures of the stand column and the transverse beam are simplified, the installation efficiency is improved, the aesthetic property of curtain wall indoor surfaces is ensured, as the connection structure can be disassembled, the connection structure is convenient to maintain, safe and reliable, and due to adoption of the closed sectional material, the material cost is saved.
