The method comprises the following steps: on the basis of a black box theory, by utilizing the self-closed landform condition of the karst depression and the particularity of the karst depression as a key node for collecting surface water and converting the surface water into underground water, a leakage channel and a surrounding leakage zone are taken as a controllable black box, and a treatment scheme is put forward according to the boundary condition of the black box through an investigation means, and a leakage channel of the black box can be blocked through fine construction; the connection between the depression and an underground river is blocked, so that the water body to be converted into underground water is reserved in the depression of the ground surface, and the purpose of blocking the karst depression to form the reservoir can be achieved due to the natural sealing of the depression terrain, so that a natural lake is formed. The problems that water resources in the karst region are prone to loss, and large-scale industrial water and domestic water of residents cannot be met are solved.
