The invention provides a design method and system of a protective air-tight door with an aluminum honeycomb sandwich structure . The method comprises the following steps that an equivalent static load of the aluminum honeycomb sandwich structure protective air-tight door under the direct action of nuclear explosion air shock wave overpressure is determined; based on a preset surface board thickness model and a preset core material thickness model, according to the surface board material, the core material material and the equivalent static load, the thickness value range of the surface board and the thickness value range of the core material are determined; based on a preset bending stress model, the thickness of the surface board is adjusted until the bending stress of the surface board is smaller than or equal to the allowable bending stress of the surface board; and based on a preset shear stress model, the thickness of the core material is adjusted until the shear stress of the core material is smaller than or equal to the allowable shear stress of the core material, and based on a preset deflection model, the door leaf deformation amount of the honeycomb sandwich structure protective air-tight door is determined, and the thickness of the core material is adjusted again or the material of the core material is adjusted according to the door leaf deformation amount.
