The invention provides a flood gate capable of being quickly locked and sensing the water level and an operation method. The flood gate comprises a basic door frame structure used for installing the whole gate; a gate structure is hinged to the basic door frame structure through a hinge; a lock installation base is fixedly installed on the gate structure; a horizontally-arranged transmission rack is installed on the lock installation base in a sliding fit mode; the transmission rack is connected with a power device used for driving the transmission rack to horizontally move; the transmission rack is connected with a plurality of sets of vertical spring bolt assemblies in a matched mode and drives the vertical spring bolt assemblies to achieve vertical locking; the vertical spring bolt assemblies are matched with a sliding limiting structure installed on the lock installation base; and a transverse spring bolt rod is arranged on the vertical spring bolt assembly located on the outermost side. The flood gate can be quickly put into use, flood is blocked on the downstream side, and therefore safe operation of an underground plant of a hydropower station is guaranteed.
