The invention relates to a prefabricated building transfer floor reserved steel bar positioning system and a construction method, and the positioning system comprises a positioning plate which is provided with a positioning hole corresponding to the position of a reserved steel bar; the fixing pieces are connected to the two opposite sides of the positioning plate; and the prisms are arranged on two opposite sides of the positioning plate. According to the positioning system, the positioning holes in the positioning plate are adopted for positioning the reserved steel bars, the position of the positioning plate can be accurately measured through the two prisms arranged on the positioning plate, and after it is guaranteed that the position of the positioning plate is accurately positioned, the position precision of the reserved steel bars can be guaranteed; the positioning plate is fixed to the formwork of the transfer layer through the fixing piece arranged on the positioning plate, disturbance to the position of the reserved steel bar caused by concrete pouring of the transfer layer can be avoided, and the position precision of the reserved steel bar is further ensured.
