The invention discloses an inflation system and an inflation method of an overwater trestle. The inflation system comprises a rubber air bag part, an inflation part and an electrical control part, the rubber air bag part comprises more than two side air bags, and the side air bags are used for providing buoyancy for the bridge span on water; each side air bag is provided with a ventilation pipe, and each ventilation pipe is provided with an electromagnetic ball valve. The inflating part comprises a vehicle-mounted inflating part and a bridge span inflating part; the vehicle-mounted inflating part is used for quickly inflating and deflating the side air bags; the bridge span inflation part is used for supplementing air to the side air bags; and the electrical control part is used for controlling the opening and closing of all the electromagnetic ball valves according to the air pressure value in the side air bag so as to inflate, deflate or supplement air to the side air bag. The multiple side air bags can be inflated at the same time before the trestle flashes, the multiple side air bags are exhausted at the same time before the trestle is loaded on the carrier loader, and when the trestle sails on the water, air is supplemented after the side air bags leak air and the air pressure is reduced.
