The invention provides a dam-penetrating box culvert anti-seepage structure and a construction method of the dam-penetrating box culvert anti-seepage structure. A closure ring integrally and annularly wraps the periphery of a box culvert structure and is close to the dam corner side of the upstream face of the box culvert structure; concrete backfill layers are backfilled on the side walls on the two opposite sides of the box culvert structure and the excavation faces of the side slopes; the grid backfill layers are backfilled above the corresponding concrete backfill layers and are located in a trapezoidal space defined by the side walls on the two opposite sides of the box culvert structure, the concrete backfill layers, the slope excavation faces and the top face of the box culvert structure; and the clay backfill layer is backfilled on the grating backfill layer, the box culvert structure and the closure ring. According to the structure, the influence of seepage on the dike can be reduced, the backfilling quality is high, post-construction settlement and differential settlement can be effectively reduced, and the construction quality is guaranteed.
