The invention provides a prefabricated box culvert water-stop belt fixing device and a construction method, the prefabricated box culvert water-stop belt fixing device is used for fixing a water-stop belt to the end of a prefabricated box culvert to be poured, and the prefabricated box culvert water-stop belt fixing device comprises an end formwork, a clamp and a fixing support; the end formwork is formed by splicing multiple sections of structured formwork units, and the overall outline of the end formwork is attached to the outline shape of the end face of the prefabricated box culvert. An inwards-sunken mounting groove is formed in the end formwork, and the mounting groove is located in the middle of the end formwork at any section of the extension path of the end formwork. The clamp is formed by splicing multiple sections of structured clamp units, the clamp can cover the whole extending path of the mounting groove, the clamp can be oppositely mounted in the mounting groove and fix the water stop belt, and a nut is welded to the side, close to the center of the mounting groove, of the clamp; the fixing support is used for fixing the end formwork to the end of the prefabricated box culvert to be poured. The construction cost can be saved, the construction efficiency and the turnover efficiency are high, and the construction quality is good.
