The invention provides a positioning device and a positioning method for a prestressed column fixed end anchorage device and a stand column main reinforcement. The positioning device comprises a positioning plate, an adjusting mechanism and a horizontal detection mechanism, wherein the positioning plate is provided with a first positioning hole for a fixed end anchorage device to penetrate through and a second positioning hole for a stand column main reinforcement to penetrate through; one ends of the adjusting mechanisms are arranged on the positioning plate, the other ends of the adjusting mechanisms are used for being arranged on the foundation pit support, the multiple adjusting mechanisms are arranged around the positioning plate, and the length of the adjusting mechanisms located between the positioning plate and the foundation pit support can be adjusted. The levelness of the positioning plate is adjusted by adjusting the length of the adjusting mechanism located between the positioning plate and the foundation pit support. The levelness detection mechanism is arranged on the positioning plate and can detect the levelness of the positioning plate. The positioning device can improve the perpendicularity of the anchorage device at the fixed end and the perpendicularity of the main rib of the stand column.
