The invention discloses a plant urease curing agent and application of the plant urease curing agent in an impermeable lining of a refuse landfill. The plant urease curing agent comprises plant urease, soluble calcium salt, urea, an adhesive and water. The mass ratio of the plant urease to the soluble calcium salt to the urea to the adhesive to the water is 10:5-10:5-30:1-2:200-400. A plant urease curing agent is mixed with clay according to the liquid-solid volume ratio of 110 L/m3, laid layer by layer and compacted on the bottom layer and the periphery of a refuse landfill to form an anti-seepage lining, the strength of the anti-seepage lining is improved by 1.75-2.53 times, the permeation heavy metal ions are reduced to 27-65%, the permeability coefficient is reduced to 1*10-7 or below, and thereby migration and permeation of the heavy metal ions are effectively reduced.
