Ceramic hydroxyapatite for chromatography as well as preparation method and application thereof
The invention provides ceramic hydroxyapatite for chromatography and a preparation method and application thereof, the preparation method comprises the following steps: 1) reacting calcium hydroxide with phosphoric acid, and adding an alkalizer to terminate the reaction to obtain slurry; 2) uniformly mixing the slurry with an additive to obtain mixed slurry; 3) performing spray granulation on the mixed slurry to obtain granules; and (4) sintering the granules, so as to obtain the ceramic hydroxyapatite for chromatography. The preparation process is simple, has no special requirements on equipment, is good in batch stability of products, and is easy to industrialize; the prepared ceramic hydroxyapatite particles for chromatography are uniform in sphericity, strong in mechanical property, large in specific surface area and good in binding specificity, and can be used as a chromatographic column filler for purifying protein and/or nucleic acid.
本发明提供一种层析用陶瓷羟基磷灰石及其制备方法、用途,所述制备方法包括以下步骤:1)将氢氧化钙和磷酸反应,加入碱化剂终止反应,得浆料;2)将所述浆料与添加剂混合均匀,得混合浆料;3)将所述混合浆料喷雾造粒,得颗粒料;4)烧结所述颗粒料,即制得所述层析用陶瓷羟基磷灰石。本发明制备工艺简单,对设备无特殊要求,产品批次稳定性好,易于产业化;制得的层析用陶瓷羟基磷灰石颗粒球形均匀,机械性能强,比表面积大,结合特异性好,可以作为层析柱填料用于纯化蛋白和/或核酸。
