The invention relates to a preparation method of an asphalt mixture, which is characterized by comprising the steps of preheating an asphalt mixture stirring container to 150-170 DEG C, adding 10-15 parts by weight of asphalt and 3-5 parts by weight of rubber particles, and quickly stirring at the stirring speed of 350-450 r/min for 20-30 minutes; and preheating 70-80 parts of treated aggregate and 15-20 parts of mineral powder to 150-170 DEG C, adding into a stirring container, stirring for 10-20 minutes, heating the stirred material to 180-190 DEG C, adding 2-3 parts of glass fibers, and continuously stirring for 5-10 minutes to obtain the asphalt mixture. The anti-stripping agent can reduce the contact angle of the asphalt on the surface of the aggregate and improve the dispersion degree of the asphalt on the surface of the aggregate. Water is not easy to retain between the aggregate and the asphalt film, so that the influence of water on the mixture is reduced. The anti-stripping agent is stable and cannot be damaged by water to be dissolved or invalid.
