The invention relates to the technical field of immersed tubes, in particular to an assembly line prefabrication system and method of an immersed tube. The prefabrication system comprises a steel bar machining area, a steel bar binding area, a pile platform and a steel buoyancy tank which are sequentially arranged, the steel bar machining area is located on the land, the steel bar binding area is located on the land or the pile platform, the pile platform is located at the junction of water and land, the steel buoyancy tank is arranged in the water area, the steel buoyancy tank is used for pouring, outfitting and launching of the immersed tube, the pile platform is in butt joint with the steel buoyancy tank, and land equipment enters and exits from the steel buoyancy tank through the pile platform. The assembly line prefabrication system of the immersed tube is a factory system combining an overwater platform and a shore platform, a pouring area and an outfitting area of a factory are combined into one buoyancy tank platform area which serves as the shared area for pouring and outfitting of the immersed tube; and a full-length inner mold process is adopted for pouring to improve the pouring progress, an outfitting sealing door adopts a sealing door-steel beam integrated structure and a sealing door pull fitting installation process, so that the outfitting installation progress is improved, the total construction period of pouring and outfitting is approximate to the steel bar binding construction period, and pouring and outfitting are just matched to form assembly line production.
