The invention discloses a construction method of a prefabricated pier column vertical rotation formwork, which belongs to the technical field of bridge construction and comprises the following steps: assembling a formwork mechanism on a formwork assembling pedestal, fixing one end of the formwork mechanism on a formwork fixing beam after the assembling is finished, connecting the formwork fixing beam to a vertical rotation pedestal through a rotating shaft, and fixing the formwork mechanism on the vertical rotation pedestal through the rotating shaft; a temporary hoisting block is installed at the other end of the formwork mechanism and used for hoisting the formwork mechanism; a lifting device is installed on the temporary lifting block, slow lifting is conducted, so that the formwork mechanism drives the formwork fixing beam to clockwise rotate by 90 degrees around the rotating shaft, and the formwork fixing beam falls on the vertical rotating pedestal; and after the formwork mechanism is vertically placed and stabilized, bolts between the formwork mechanism and the formwork fixing beam are dismantled, the formwork mechanism is integrally hoisted to a pouring pedestal to be fixed, the temporary hoisting blocks are dismantled, and concrete pouring construction is conducted. The advantages of horizontal construction and vertical construction are ingeniously combined, the safety during formwork assembly is improved, and the apparent quality of concrete is guaranteed.
