The invention discloses a laboratory layout structure of a secondary protective barrier of a three-level biosafety laboratory, which comprises a negative pressure protection area and a positive pressure auxiliary protection area, and is characterized in that the negative pressure protection area comprises a BSL-3 core experiment area, an ABSL-3 core experiment area, a buffer room, an animal quarantine room, an article temporary storage room, an animal residue storage room and a second personnel protection area; the positive pressure auxiliary protection area comprises an animal front chamber, a first cleaning preparation room, a second cleaning preparation room, a protection corridor and a first personnel protection area; and a buffer room is arranged between the BSL-3 core experiment area and the ABSL-3 core experiment area for communication. The protection corridor surrounds the negative pressure protection area in a three-face mode. The whole laboratory layout structure is provided with a BSL-3 three-level microbial biosafety laboratory secondary protection barrier and an ABSL-3 three-level animal biosafety laboratory secondary protection barrier at the same time, and it is totally guaranteed that harmful biological factors in the biosafety laboratory cannot escape out of the laboratory; and the cleanliness of the laboratory is ensured to meet the national standard cleanliness requirement on the whole.
