The invention provides a bridge deck slab pre-tightening system and a bridge deck slab pre-tightening construction method.The bridge deck slab pre-tightening system comprises a steel cross beam, a bridge deck slab pre-tightening device and a bridge deck slab pre-tightening device, the outer sides of the bridge deck slabs located at the two ends of the steel cross beam abut against the check blocks located at the two ends of the steel cross beam, and the length direction of the bridge deck slabs is the same as the length direction of the steel cross beam; the pre-tightening jacks are arranged between the adjacent bridge deck slabs and used for applying pre-tightening force to the bridge deck slabs in the length direction of the bridge deck slabs so as to enable the bridge deck slabs to be compressed in the length direction, and when the pre-tightening force applied to the bridge deck slabs by the pre-tightening jacks reaches a target value, the pre-tightening jacks are used for pre-tightening the bridge deck slabs in the length direction of the bridge deck slabs. The jacking force is continuously kept so that the bridge deck slab can be kept in a pre-tightening state; and the two isolation templates are arranged on the two sides of the pre-tightening jack, so that an isolation space is formed, and the pre-tightening jack is isolated from the outside. The bridge deck slab pre-tightening system has good economical efficiency and safety.
