The invention discloses a positioning device and method for construction of a steel pipe column of a subway station through a cover and excavation top-down method. The positioning device comprises a fixed track beam, a transverse moving track beam, a longitudinal moving track beam, a positioning pulley assembly and a positioning instrument. The two fixed track beams are parallel and fixed at intervals, and the pair of transverse moving track beams are installed on the two fixed track beams through the positioning pulley assemblies and are perpendicular to each other. The pair of longitudinal moving track beams is installed on the two transverse moving track beams through the positioning pulley assemblies and is perpendicular to the two transverse moving track beams. Meanwhile, the positioning pulley assembly in the transverse moving track beam is located on the inner side of the positioning pulley assembly in the fixed track beam; the positioning instrument is placed on the pile top or fixed above the steel pipe column; the position indicator projects towards the center of the steel pipe column, the position of the steel pipe column is adjusted by moving the transverse moving track beam and the longitudinal moving track beam, and the pile center coincides with the steel pipe column center. According to the invention, the steel pipe column can be quickly and conveniently positioned.
