The invention provides a cooling tower cooling water supply system and method with pure water replenishment, the cooling tower cooling water supply system comprises a cooling tower, a cooling tower cooling water supply pipeline, a tap water supply unit and a pure water supply unit, and the cooling tower is connected with the tap water supply unit and the pure water supply unit through the cooling tower cooling water supply pipeline; the tap water supply unit comprises a tap water pipe which is connected with a tap water room and a cooling tower cooling water supply pipeline, and a stop valve is mounted on the tap water pipe; the pure water supply unit comprises a pure water supply pipeline, a pure water supply pipeline valve, a closed water tank and a pure water room, the cooling tower cooling water supply pipeline, the closed water tank and the pure water room are sequentially connected through the pure water supply pipeline, and the pure water supply pipeline is provided with the pure water supply pipeline valve; the closed water tank is arranged on the lower portion of the cooling tower so as to collect cooling water falling from the cooling tower, and pure water is adopted for assisting water supply, so that the operation cost of the cooling tower is reduced.
