The invention provides an automatic measuring device and method for open caisson construction positioning, and relates to the technical field of open caisson construction. The open caisson construction positioning automatic measuring device comprises a foundation, a foundation pit, a computer, a linear displacement sensor, an open caisson body and four sets of unmanned aerial vehicles. According to the method, the four unmanned aerial vehicles hover above the open caisson body to collect real-time height information, the instantaneous position deviation, the inclination deviation and the settlement amount in the sinking process of the open caisson body are calculated through a computer program, and adjustment of a construction scheme during sinking of the open caisson body is guided according to a result; and the center of the open caisson and the reserved hole are ensured to sink to preset positions. Only the overhead range of the open caisson is occupied in open caisson construction control, the requirement for the foundation range is greatly reduced, continuous power supply is conducted on the unmanned aerial vehicle through the power supply box and the cable, the unmanned aerial vehicle can obtain longer endurance time, and the target disc serving as the measuring basis can be dismantled after construction is completed and can be reused on a subsequent new open caisson construction site.
