The invention provides a building structure hole plugging device and a construction method. The building structure hole plugging device comprises a plugging assembly used for plugging a hole; a reference piece used for aligning the upper end surface of the hole; a limiting piece used for limiting the position of the reference piece; and a connecting assembly sequentially penetrating through the limiting piece, the reference piece, the hole and the plugging assembly and enabling the limiting piece, the reference piece and the plugging assembly to be connected; wherein the plugging assembly canpenetrate through the hole along the diagonal line of the hole and is horizontally placed, and the plane of the plugging assembly makes contact with the lower end face of the hole. The connecting assembly can freely rotate relative to the plugging assembly and penetrates through the hole in height; and the distance between the plugging assembly and the reference piece is the height of the hole. The device is simple in structure, easy to operate, capable of being repeatedly used, low in machining cost, good in reliability, free of influences of the floor thickness and the hole shape and size, wide in application range and high in practicability.
