The invention discloses a construction method of a prestressed anti-floating anchor rod, which comprises the following steps of: constructing an anchor backing plate, so that the anchor backing plate is sleeved on the anchor rod and is positioned at a position above a cushion layer of a foundation slab; binding a reinforcement cage of the foundation slab, so that reinforcements of the reinforcement cage are staggered from the anchor backing plate; the foundation slab is poured, the bottom of the anchor bearing plate and the foundation slab are poured together, and the space above the top of the anchor bearing plate is reserved with the periphery of the anchor bearing plate as the boundary; the anchor rod is subjected to prestress tensioning in the reserved space; the reserved space is backfilled, and the construction period is shortened through the construction method.
