The invention relates to the technical field of oil field well cementation, and discloses a preparation method and application of a high-temperature-resistant fluid loss agent and cement paste. The method comprises the following steps: contacting 2-acrylamido-2-methylpropanesulfonic acid, an unsaturated monomer, a silane coupling agent, an initiator, a reducing agent and graphene oxide for reaction to obtain the high-temperature-resistant fluid loss agent, the unsaturated monomer is selected from one or more of N-vinyl pyrrolidone, acrylamide, acrylic acid, methacrylic acid, vinyl sulfonic acid or vinyl sulfonate, vinyl phosphoric acid or vinyl phosphate, maleic acid or maleic anhydride, itaconic acid, fumaric acid, styrene sulfonate, allyl sulfonate and hydroxyethyl methylacrylate. The high-temperature-resistant fluid loss agent provided by the invention is good in high-temperature resistance and high in mechanical strength, and is particularly suitable for well cementation engineering of deep and ultra-deep oil and gas wells.
