The preparation method comprises the following steps: mixing gypsum powder and dispersion liquid according to a certain solid-to-liquid ratio, and fully stirring to obtain first slurry; adding a certain amount of a crystal phase transition agent and a crystal morphology guiding agent into the first slurry, and stirring and reacting for a period of time to obtain second slurry; adding an antibacterial precursor into the second slurry, regulating the pH value of the slurry to be acidic, and stirring for reaction to obtain third slurry; and filtering, washing, drying and crushing the third slurry to obtain the gypsum-based antibacterial material. The relative texture coefficient of the 204 crystal face of the gypsum-based antibacterial material is greater than 0.1. According to the invention, the alpha-CaSO4. 5H2O material with intrinsic antibacterial activity is prepared through gypsum crystal transformation and in-situ structure regulation and control.
