The invention discloses a cable-stayed bridge steel main tower section positioning device and positioning method. The cable-stayed bridge steel main tower section positioning device comprises: a supporting steel plate welded and fixed to a steel main tower wall plate; four main steel plates supported on the supporting steel plate and arranged around steel main tower; a horizontal jack set and a vertical jack set which are installed on the main steel plates, wherein the horizontal relative positions of two adjacent sections of the steel main tower are adjusted through the horizontal jack set; the vertical relative positions of every two adjacent sections of the steel main tower are adjusted through the corresponding vertical jack set. The positioning device can be repeatedly used, and is easy to mount and dismount, more stable, accurate in positioning and rapid in positioning; the device has very high flexibility and adaptability, and is good in applicability; and all parts of the positioning device are made of the same steel material as the steel main tower, mounting and dismounting are easy and convenient, and modification is easy.
