The invention relates to a prefabricated segmental beam axis measuring and positioning device and a measuring method, and the device comprises a pre-embedded assembly which is used for being pre-embedded on a prefabricated segmental beam in the transverse bridge direction, and the two ends of the pre-embedded assembly are located on the two sides of the axis of the prefabricated segmental beam respectively; a connecting piece; scales are arranged on the transverse rod; the cross rod is parallel to the embedded assembly and is detachably connected with the embedded assembly through a connecting piece; and the positioning indicating piece is movably arranged on the cross rod, indicates the scale value of the positioning indicating piece and is used for providing a mark point of the total station. In the factory-leaving section, the transverse rod and the positioning indicating piece are installed on the pre-embedded assembly through the connecting piece, and then factory-leaving axis point scales corresponding to the axis points of the prefabricated segmental beam are obtained through the total station; when the prefabricated segmental beam is installed on the bridge pier, only the positioning indicating piece needs to be moved to the factory axis point scale, so that when the prefabricated segmental beam is erected, the axis point can be obtained without measurement, the influence of disappearance of the marking point is avoided, and the reflector plate is prevented from being supported.
