The invention provides a pre-stressed anchor rod tensioning and locking structure which comprises a concrete cushion layer laid below a basement bottom plate, a steel plate is arranged on the concrete cushion layer, a stirrup steel plate for supporting a hydraulic jack is fixed to the steel plate, a pre-stressed anchor rod penetrates through the concrete cushion layer, the steel plate and the stirrup steel plate, and a first anchoring plate is fixed to the top end of the pre-stressed anchor rod. The hydraulic jack is located between the first anchoring plate and the stirrup steel plate, the prestressed anchor rod tensioning and locking structure further comprises a nut used for locking the prestressed anchor rod, and the nut is tightly attached to the steel plate. The invention further provides a pre-stressed anchor rod tensioning and locking method.
