The invention provides a fully-prefabricated flat slab system connected through prestress and a construction method of the fully-prefabricated flat slab system. The fully-prefabricated flat slab system comprises a fully-prefabricated floor slab and a prefabricated column supported on the lower surface of the fully-prefabricated floor slab. Column longitudinal bars extend out of the top faces of the prefabricated columns, and column top brackets are arranged on the tops of the prefabricated columns. An anchoring screw extending outwards is arranged on the top surface of the column top bracket; prestressed reinforcements are arranged at the upper part of the fully-prefabricated floor; a pre-embedded metal corrugated pipe is arranged in the middle of the fully-prefabricated floor slab, and screw through holes vertically penetrating in the thickness direction of the fully-prefabricated floor slab are formed in the periphery of the pre-embedded metal corrugated pipe. Prestressed ducts are formed in the four edges of the fully-prefabricated floor slab; column longitudinal bars on the top faces of the prefabricated columns are connected with the embedded metal corrugated pipes in an inserted mode and anchored through grouting. The anchoring screw rod is connected with the screw rod through hole in an inserting manner and is fixed through a nut and a gasket; the prestressed tendons are arranged in the prestressed hole channels of the two fully-prefabricated floor slabs in a penetrating mode. The invention can solve the technical problems of complicated mounting and dismounting work, large wet work workload and large labor demand of the flat slab construction site support template.
