A foundation pit is excavated, concrete is poured into the foundation pit to form a cushion layer, and positioning steel bars are pre-buried; angle iron is welded to the positioning steel bars, a base formwork is built between the angle iron and the cushion layer, and concrete is poured into the base formwork to form a base; laying a steel plate on the base, and welding the steel plate with the angle iron, so that the steel plate and the angle iron jointly define a containing groove; the rubber pipe is plugged into the containing groove, the axial direction of the rubber pipe is the same as the extending direction of the containing groove, and part of the rubber pipe is arranged outside the containing groove; and a box girder formwork is built outside the base, the rubber pipe is pressed into the containing groove through the box girder formwork, and concrete is poured into the box girder formwork to form the box girder. According to the construction method of the prefabricated box girder, the slurry leakage condition can be effectively prevented, the bottom of the box girder is prevented from generating a honeycomb pitted surface, the quality and attractiveness of the box girder are guaranteed, and therefore the labor and material cost generated by finished product modification is avoided.
