The medium-temperature yeast fermentation room comprises a yeast room and a yeast frame located in the yeast room, the yeast room is of a tunnel type structure, a yeast room door is arranged at one end of the yeast room, a rear wall is arranged at the other end of the yeast room, a lighting window is arranged in the middle of the rear wall, and a moisture removal window is arranged at the top of the rear wall; and the koji frame is of a multi-layer bracket structure. By adopting the tunnel type koji room structure and matching with the multilayer koji frame to place koji blanks, the civil engineering utilization rate of the koji room can be effectively improved, the multilayer overlapped koji blanks can better gather the fermentation temperature, and in the fermentation process, the koji room door at one end of the koji room and the lighting window and the moisture removal window at the other end of the koji room are utilized to realize air convection, so that the fermentation efficiency is improved. Therefore, a heat-preservation and moisture-preservation micro-aerobic environment condition suitable for the growth of starter propagation microorganisms is created, a traditional heat-preservation and moisture-preservation covering is abandoned, and the sensory index and the physicochemical index of the yeast are remarkably improved while the quality of the yeast is guaranteed.
