The invention relates to gypsum-based seal mortar for through-wall holes and a preparation method of the gypsum-based seal mortar. The gypsum-based seal mortar is prepared from components in parts by weight as follows: 100 parts of flue gas desulfurization gypsum, 30-250 parts of an active admixture, 0.01-10 parts of a water reducer, 0.1-50 parts of an activator, 0-10 parts of an adjusting admixture, 0-20 parts of a binder, 0-10 parts of a water repellent, 0.01-30 parts of a plastic retaining agent, 0.01-10 parts of starch ether and 30-300 parts of quartz sand. Compared with the prior art, the gypsum-based seal mortar for through-wall holes and the preparation method have the advantages as follows: the workability is good; the operable time is long; newly stirred mortar has good plasticity, adhesive force and falling and skidding resistance; a hole can be extruded, injected and sealed with a caulking gun or a caulking machine, a flowing phenomenon is not caused even when a large hole is filled, the hole can be fully filled to the deep position and sufficiently sealed, the sealing material plays a role sufficiently, and the like. A large amount of industrial waste gypsum is used, the workability of the material can be improved, the material cost can be reduced, and a novel way is provided for application of the industrial waste gypsum.
