The invention provides an aluminum oxide ceramic substrate and a preparation method and application thereof, and belongs to the technical field of ceramic circuit substrates. The preparation method of the aluminum oxide ceramic substrate comprises the following steps: mixing an abrasive and a grinding solvent, carrying out first-time ball milling, mixing an obtained first mixed solution with a binder, carrying out second-time ball milling, adding a defoaming agent into an obtained second mixed solution, and carrying out defoaming treatment and ageing treatment to obtain slurry; wherein the grinding material is alumina powder and a dispersing agent, and the dispersing agent is polyvinyl acetate and sodium silicate; carrying out tape casting on the obtained slurry on a tape casting machine to form an aluminum oxide green body, and then preparing an aluminum oxide ceramic green sheet; and sintering the obtained aluminum oxide ceramic blank sheet to obtain the aluminum oxide ceramic substrate. According to the preparation method of the aluminum oxide ceramic substrate, the toughness and the strength of the aluminum oxide green body can be remarkably improved.
