The invention discloses a cement plug plugging method under an oil-based drilling fluid condition and cement paste, and the method comprises the following steps: a drilling tool is put down to a well bottom or a leakage layer, and a pre-positioned spacer fluid is pumped at a first preset pumping displacement; cement paste is pumped with the second preset pump injection displacement; if the front spacer fluid flows out of the water hole of the drilling tool and starts to return upwards by a first preset height, drilling is conducted by one stand column height without stopping the pump; pumping the rear spacer fluid at a third preset pumping displacement; the drilling tool is kept immobile, and the drilling fluid is pumped with the fourth preset pumping displacement; judging whether the preposed spacer fluid flows out of the water hole of the drilling tool and begins to upwards return to the annulus or not; if yes, after the front spacer fluid upwards returns to the first preset height, the drill is pulled out by the height of one stand column without stopping the pump; if not, pulling out the hole to a second preset height above the cement surface; well closing and squeezing injection are conducted, and spacer fluid arranged in front of the bottom of the cement paste and cement paste with the sixth preset volume are squeezed into the leakage stratum; and according to the determined initial setting time of the cement paste, well shut-in squeezing injection is conducted with the preset squeezing injection displacement, and the total squeezing injection amount of the cement paste is determined.
